High-Throughput Systematic Evolution of Ligands by EXponential Enrichment (HT-SELEX) assays were carried out for 392 transcription factors (TFs). A total of 1585 individual HT-SELEX experiments were performed, each of which was performed for three or four cycles. The data generated from these assays and the experimental details are accessible here. ## Metadata ## Experimental details can be downloaded from the website under the HT-SELEX heading - "Metadata(.xlsx)" The Excel file contains 5 individual spreadsheets: (1) "README": Contains column descriptions for the remaining 4 spreadsheets (2) "HT_Overview_TF": Contains, for each of the 392 TFs, the number of HT-SELEX experiments performed and how many were successful. (3) "HT_Overview_DBD": Contains, for each construct design across each TF, the number of HT-SELEX experiments performed and how many were successful. This differs from "HT_Overview_TF" in that it differentiates between experiments performed for the same TF but with full-length constructs vs. DBD constructs, which contain only the DNA-binding region of the protein, or a portion of the DNA-binding region. (4) "HT_Experiment_Info": Contains a row for each individual experiment and provides further details. (5) "Control_Info": Contains a row for each individual control experiment. ## Raw Data ## HT-SELEX data can be downloaded from the website under the HT-SELEX heading - "Raw: All Raw data(.tar.gz)" This directory contains 5 subdirectories: The subdirectories "Cycle1", "Cycle2", "Cycle3" and "Cycle4" contain 1581, 1583, 1585, and 1081 FASTQ files, respectively. Each file corresponds to a single cycle from a single experiment and contains the trimmed sequencing reads. Files are distributed across the four subdirectories according to their respective SELEX cycle. Details for a given file can be found by looking up the file name in the "HT_Experiment_Info" spreadsheet found within the HT-SELEX Metadata Excel file described above. The "Control" subdirectory contains 99 FASTQ files containing trimmed reads. Three are bead only (beadOnly*) experiments and the remaining 96 are unselected library controls, serving as cycle 0. These can be matched up to experiments using the "HT_Experiment_Info" and "Control_Info" sheets in the Excel file.
